I have an off-day on Saturday while traveling on business.
If you only had one day to spend in Miami (or surrounding area), what would you recommend?

Take an all-day Everglades tour and spend the night hanging out in South Beach.
I've done this tour, from the Ft. Meyer side (they do a similar tour from the Miami/Lauderdale side):

And found it entirely enjoyable - no "Eco" preachiness that I was able to discern.

If you like the beach, Key Biscayne is nice. The "other" Keys (like Key Largo) are about a 2+ hour drive, and not really worth the trip unless you are planning on diving or fishing.
South Beach at night is cool. Never been there during the day though.

I have a friend with a flat on the beach at the Trump Hollywood Tower and a New Mercedes 450 SL.
