Can I pop an external into my mac and just mosey on?

The 500 gigger in my white iMac C2D 2.16 (OS 10.5.5) isn't cuting it any more.
I recently bought a 1 TB Seagate and popped it into an external USB case, and have been using it for a backup drive.

Can I take that drive out of the USB case and swap it into the iMac and continue on with life, or is there something specific about the formatting while in an external case that might prevent that?

I know it's kind of a dumb question but I do this kind of stuff very rarely any more and I have some vague memory of it being problematic in the past (maybe back in the SCSI age, I don't know).

(edit-- also, any reason I wouldn't want a 1 TB drive in a late 2006 iMac?)

It has to be GUID partition scheme (or whatever it is called) to boot an Intel Mac. Check the Disk Utility app, specifically in the partition area, and erase the external drive first.

After that, do a Carbon Copy Clone to it, copying everything over from the internal drive, and it should run just fine while inside the machine.

As long as you formatted the external as HFS+ and GUID when you first used it you should be good to go.

As long as it has the same connector (IDE, I think), you'll be fine. My suggestion on the easiest swap solution: use Carbon Copy Cloner to clone your external drive from the internal one (and make sure it is bootable), and then just do the swap. You'll be up and running on the new drive, with all of your files immediately after you install the new drive. You should, however, repair permissions once you boot with the new drive, and make sure to set it as your default boot-up drive in System Preferences-->Startup Disk.

There was a 750GB IDE (PATA) drive but it's not very common.

Yup, SATA all the way, no IDE in the picture.
As stated, it's my backup drive-- already CCC'd to hell and back.
I have a new TB-er (I think the exact same model as the external) that's going to become the new backup drive. Not willing to test the new one inside the iMac when I have a good feeling about the old one (if you've ever broke open a white Intel iMac you'll understand).

How exactly does one confirm that a hard drive was formatted with GUID?

In disk utility, click on the drive it will show at the bottom of the window what it was formatted as.

Did that, it doesn't, except "Mac OS Extended (Journaled)."

You need to highlight the top level drive name - not the sub level volume name
