Why do I have to keep deleting and adding my printer?

I have a Brother 5340D (non-wireless) printer that used to work flawlessly with my setup. I have it hooked via a USB cable to an old Airport Express. It uses to be easy peasy to print using my MBP. However for the past month or so, I try to print something and it doesn't print. I go into the Brother printer software, and it tells me the printer is "offline". The only way to fix it seems to be to go into the system preferences, delete the printer, and then add it again.

Does anyone know what gives here?

Hard to say. Are you using the native Brother driver or Airprint?

I'm using the Brother driver...

It doesn't indicate whether it's Airprint under the device name. It indicates it at the end of the "kind" line in the main Printers & Scanners window.

You should ensure that you are using the Brother CUPS driver.

...You could try resetting the printing system: Control-click on the left side of the Printers & Scanners window and select "Reset printing system..." from the contextual menu.

....the Sister driver might work better....if adventurous, the Sista' Driver takes it to another level......

I have a vague memory of giving the AE a static IP to solve a similar in the past.

What btfc said.... had similar setup before downsizing a few years ago, w/ the AE mostly hosting a more frequently used med-high end Canon inkjet photo printer (after hosting a more seldom used HP LJ6MP). Going static stopped a whole world-o-hurt.... well, until one of the ports on the switch handling a handful of networked devices went on the fritz, causing a nuther-world-o-hurt (w/ a $20, new switch solution to stop $10K worth of agita, but that's a story for another time...).

My best guess is that you have a dynamic IP on the printer and it keeps moving as other devices join your network.
