Run DiskInventoryX to see what's hogging your drive. It will show a visual representation of the contents as blocks so that you can see which files or directories are guilty. I often find huge mistakenly saved files that way.

Make sure that your backups are up to date.
Then run this in the Terminal:
sudo tmutil thinlocalsnapshots / 1000000000 4
(This is a good command to run if you delete a lot of files and you don't see your drive-capacity increase. It purges storage reserved for local snapshots left behind from Time Machine and OS updates.)

Fyi, dabbling with DiskInventory x now.
Did a fresh full back up last night and the source disk that now shows Used: 860.2 Free: 134.5 and the back up shows Used: 550.1 Free: 449.9.
